Description

Thiopental Sodium for Injection

Healthy Inc is a specialized global supplier and exporter of essential anesthesia and neurocritical care therapies. We provide high-purity Thiopental Sodium for Injection (500 mg / 1000 mg), manufactured in WHO–GMP certified sterile lyophilization facilities. This “Ultra-Short-Acting Barbiturate” is a mandatory export to neurosurgical centers, emergency trauma units, and government health ministries in Africa, LATAM, and Southeast Asia, serving as a reliable gold standard for the induction of general anesthesia and the emergency management of refractory status epilepticus.

Product Overview

This formulation contains Thiopental Sodium (also known as Thiopentone), a sulfur-containing thiobarbiturate derivative that rapidly depresses the central nervous system.

The “Neuro-Depressant” Specialist:

  • Mechanism (Prolonged GABA-A Opening): Thiopental binds to a specific site on the GABA-A receptor. Unlike benzodiazepines (which increase the frequency of chloride channel opening), barbiturates increase the duration that the chloride channels remain open. This massive influx of chloride ions intensely hyperpolarizes the neuron, resulting in profound, rapid unconsciousness (typically within 30 to 45 seconds).
  • Neuroprotection & Intracranial Pressure (ICP): Thiopental causes potent cerebral vasoconstriction, which drastically reduces cerebral blood flow and cerebral metabolic oxygen requirements (CMRO2). This makes it the preferred induction agent for patients with severe traumatic brain injury, brain tumors, or life-threatening elevations in intracranial pressure.
  • Status Epilepticus: Its powerful CNS-depressant effects make it a critical last-line intervention to break continuous, medication-resistant seizures (inducing a “barbiturate coma”).

Manufacturing Authority

Marketed and Distributed by Healthy Inc from WHO-GMP & ISO 9001:2015 certified units.

  • Alkaline Buffering (pH Control): Pure thiopental is unstable in water and forms an insoluble free acid. Our formulation strictly incorporates Anhydrous Sodium Carbonate during lyophilization. Upon reconstitution with Water for Injection, this creates an intensely alkaline solution (pH 10.5 to 11.0), guaranteeing the active pharmaceutical ingredient dissolves completely without lethal precipitation.
  • Nitrogen Blanketing: Barbiturates are sensitive to carbon dioxide (which lowers the pH and causes precipitation). Vials are purged and sealed strictly under a nitrogen atmosphere to preserve chemical integrity.

Therapeutic Indications (Human Use)

Indicated for use by trained anesthesia and neurocritical care personnel only:

  • General Anesthesia: Sole anesthetic agent for brief procedures (under 15 minutes) or for the rapid induction of anesthesia prior to maintenance with other agents.
  • Status Epilepticus: Emergency control of convulsive states occurring during or following inhalation anesthesia, local anesthesia, or other causes.
  • Neurosurgery: Reduction of elevated intracranial pressure (ICP) in patients with severe cerebral edema or traumatic brain injury.
  • Narcoanalysis: Used in specialized psychiatric settings for narcoanalysis or narcosynthesis.

Dosage & Administration

Recommended Dosage (Strictly as per Anesthesiologist):

  • Route: Intravenous (IV) Injection ONLY.
  • CRITICAL WARNING (Extravasation): Because the solution is highly alkaline (pH ~10.5), extravasation into surrounding tissues or accidental intra-arterial injection causes massive chemical burns, severe vasospasm, and tissue necrosis (gangrene). Must be injected securely into a large, freely running vein.
  • Induction Dose (Adults): Typically 3 to 5 mg/kg IV (as a 2.5% solution), injected slowly over 10 to 15 seconds.
  • Precipitation Hazard: DO NOT MIX with acidic drugs (such as neuromuscular blockers like Suxamethonium or Rocuronium, or analgesics) in the same syringe or IV line, as rapid, dangerous precipitation will occur. Flush IV lines thoroughly between drugs.

Safety Warnings (CRITICAL):

  • Porphyria (ABSOLUTE CONTRAINDICATION): FATAL RISK. Thiopental completely contraindicates in patients with latent or manifest porphyria, as it induces the enzyme ALA synthetase, precipitating a fatal acute porphyric crisis.
  • Airway Collapse: Causes profound respiratory depression. Apnea is common following the induction dose. Intubation and mechanical ventilation equipment must be immediately at hand.
  • Cardiovascular Depression: Causes dose-dependent myocardial depression and venodilation, leading to hypotension. Use with extreme caution in hypovolemic or shock patients.
