    • Day 2: 500 mg (300 mg base).
    • Day 3: 500 mg (300 mg base).
    • Total Dose: 2500 mg (1500 mg base) over 3 days.
  • Prophylaxis: 500 mg (300 mg base) taken once weekly on the same day of the week. Start 1-2 weeks before travel.
  • Lupus / RA: 250 mg daily (maintenance).
  • Administration: Take with food to reduce nausea and stomach pain.

Safety Warnings:

  • Retinopathy (Eye Toxicity): Long-term use (e.g., for Lupus) can cause irreversible retinal damage and blindness. Baseline and annual eye exams are mandatory.
  • Pruritus (Itching): Intense itching is a very common side effect in Black/African patients. This is not an allergy but a genetic reaction.
  • Psoriasis: Can precipitate a severe attack of psoriasis. Contraindicated in psoriatic patients.
  • Cardiotoxicity: Can cause QT prolongation and arrhythmias. Caution with other heart drugs.
