- Administration: Take after meals with plenty of water to reduce gastric irritation and ensure hydration.
Safety Warnings:
- Hypersensitivity (SJS/TEN): Rare but fatal skin reactions (Stevens-Johnson Syndrome). Discontinue immediately at the first sign of a rash. Risk is higher in patients with the HLA-B*5801 allele (common in Asian populations).
- Acute Flares: Initiation often precipitates an acute gout attack. Must be co-prescribed with NSAIDs or Colchicine for the first 3–6 months.
- Drug Interaction: Azathioprine/6-MP: Allopurinol blocks the breakdown of these drugs. If used together, the dose of Azathioprine must be reduced by 75% to prevent bone marrow toxicity.
