This formulation contains Cloxacillin (as Cloxacillin Sodium), a narrow-spectrum, bactericidal beta-lactam antibiotic.

The “Staphylococcal Shield” Specialist:

• Mechanism (Penicillinase Resistance): Many strains of Staphylococcus produce an enzyme called penicillinase (a type of beta-lactamase) that easily destroys standard penicillin G and amoxicillin. Cloxacillin is engineered with a bulky isoxazolyl side chain. This structure creates intense “steric hindrance,” physically blocking the staphylococcal penicillinase enzyme from reaching and breaking the vital beta-lactam ring, allowing the drug to successfully destroy the bacteria.
• Cell Wall Blockade: Once protected, Cloxacillin binds to specific Penicillin-Binding Proteins (PBPs) located inside the bacterial cell wall. It halts the final stage of peptidoglycan cross-linking, physically weakening the cell wall until osmotic pressure causes the bacteria to lyse (burst).
• Clinical Scope (MSSA vs. MRSA): It is highly lethal against Methicillin-Susceptible Staphylococcus aureus (MSSA) and most Streptococci. Note: It is completely ineffective against Methicillin-Resistant Staphylococcus aureus (MRSA), which has mutated its target PBPs.

Dosage & Administration

Recommended Dosage (Strictly as per Physician):

• Administration (CRITICAL TIMING): Must be taken orally on an empty stomach, at least 1 hour before or 2 hours after meals. Food significantly binds to the drug and drastically reduces its absorption and serum concentrations.
• Standard Adult Dose: 250 mg to 500 mg every 6 hours (four times daily).
• Severe Infections (Bone/Joint): Up to 1000 mg (1 g) every 6 hours.
• Pediatric Dose (Children > 20 kg): Dosed according to adult recommendations. (For children <20 kg, weight-based oral suspensions are preferred).

Safety Warnings (CRITICAL):

• Penicillin Hypersensitivity: FATAL ANAPHYLAXIS RISK. Strictly contraindicated in patients with a known history of severe hypersensitivity to any penicillin or cephalosporin antibiotic. A thorough allergy history must be taken prior to prescribing.
• Hepatotoxicity: High doses or prolonged use can cause cholestatic jaundice and hepatic impairment. Liver function tests (LFTs) should be monitored during extended therapy.
• Gastrointestinal Disruption: Like many oral beta-lactams, it can alter normal gut flora, leading to antibiotic-associated diarrhea or, rarely, Clostridium difficile-associated colitis (CDAC).
