Product Overview
This formulation contains a fixed-dose combination of Sulphadoxine (a long-acting sulfonamide) and Pyrimethamine (a diaminopyrimidine).
The “Double Blockade” Specialist:
- Synergy (1+1=10): Both drugs attack the malaria parasite’s folic acid synthesis pathway, but at different steps.
- Sulphadoxine inhibits the enzyme dihydropteroate synthase (blocking synthesis).
- Pyrimethamine inhibits the enzyme dihydrofolate reductase (blocking activation).
- Sequential Blockade: By blocking the pathway at two consecutive points, the combination is super-additive (bactericidal/parasiticidal), killing the parasite much more effectively than either drug could alone.
- IPTp Role: Because both drugs have very long half-lives, a single dose provides protection for weeks. This makes it the drug of choice for Intermittent Preventive Treatment in pregnancy (IPTp), significantly reducing low birth weight and maternal anemia in endemic zones.

Therapeutic Indications (Human Use)
Indicated for the treatment and prevention of malaria:

Dosage & Administration
Recommended Dosage (Strictly as per WHO/National Guidelines):
- Route: Oral.
- Treatment (Adults > 45kg): Single dose of 3 tablets taken all at once.
- IPTp (Pregnant Women): 3 tablets taken as a single dose. Administered at least 3 times during pregnancy (starting in 2nd trimester), with at least 1 month between doses.
- Children:
- 5-10 kg: 1/2 tablet.
- 11-20 kg: 1 tablet.
- 21-45 kg: 2 tablets.
- Administration: Take with plenty of water.
Safety Warnings:
- Sulfa Allergy (CRITICAL): Contraindicated in patients with known hypersensitivity to sulfonamides. Can cause life-threatening Stevens-Johnson Syndrome (SJS) or Toxic Epidermal Necrolysis (TEN). Stop immediately if a rash appears.
- G6PD Deficiency: Can cause severe hemolysis (anemia) in patients with G6PD deficiency.
- Infants: Contraindicated in infants under 2 months of age (risk of kernicterus).
- Folic Acid Supplements: High doses of folic acid (≥5 mg) should not be taken concomitantly as they antagonize the drug’s effect. (0.4mg standard prenatal dose is usually acceptable).
